Tips: What is a spectrophotometer?

Spectrophotometry is the absorption of light at a specific wavelength or within a range of wavelengths for qualitative or quantitative analysis of the material. The commonly used wavelength ranges are: 200-380nm UV spectrum, 380-780nm visible spectrum, 2.5-25μmInfrared spectrum. The instruments used for testing these spectrums are ultraviolet spectrophotometers, visible spectrophotometers (or colorimeter), infrared

spectrophotometers, or atomic absorption spectrophotometers.
Spectrophotometers have become conventional instruments in modern molecular biology laboratories. It is commonly used for quantification of nucleic acids, protein quantification, and bacterial growth concentrations.

The spectrophotometer instrument consists of a light source, a monochromator, a sample chamber, a detector, a signal processor, and a display and storage system.
Different light sources have their own emission spectra, so different light emitters can be used as light sources for the instrument.

  • Tungsten Emission Spectroscopy
    Tungsten light emits a spectrum of 400-760nm wavelength, which is refracted by a triangular prism and generates continuous chromatograms consisting of red orange, yellow-green, blue-violet, and violet. The spectrum can be used as a visible spectrophotometer light source.
  • Emission spectroscopy of Hydrogen (or Xenon) lamps
    Hydrogen or Xenon lamps emit wavelengths in the range of 185 to 400 nm and can be used as light sources for ultraviolet photometers.
